Can a backpack hold 40 pounds?
The weight of a backpack depends on the age of the person using it. Generally speaking, adults should not exceed 20 percent of total body mass when loading backpacks. That means a healthy person weighing 200 pounds should not carry more than 40 pounds in his or her backpack.
As a general rule, to prevent injury, a full backpack should weigh no more than 10 to 20 percent of your child's body weight.
As this shows, a 25 kg load being manually handled is considered to be a safe upper limit for the average man and 16 kg for the average woman. It's important to note, however, that this is only if the person carrying the load can keep it at knuckle height and close to their body.

If you weigh 150 pounds, you should carry no more than 15 pounds in a backpack. Here are some additional recommendations from UI Health Care occupational therapists.

21–35 liters: This is the sweet spot for most hiking and travel daypacks. They might be able to carry 10% of their bodyweight on a long hike. However, still pay attention to their posture as they hike. If your kids are leaning forward due to the strain of the weight, the pack is probably too heavy.

Carrying around a heavy backpack (a significant percentage of your own bodyweight) increases the intensity of walking as an exercise. One study found that the calorie burn during weighted walking was essentially equivalent to running (and in some cases, it was actually higher).
You have three deltoid muscles, front, middle and rear. These are the muscles you see that make shoulders look like a ball when they are large and allow you to lift something heavy over your head when you are strong. These will not get stronger or larger from carrying a backpack.
